User Guide
1. Remove a piece of leaf which shows suspect symptons.
2. Unscrew dropper from extraction bottle and place plant sample inside. Replace cap and screw on tightly - Shake bottle vigorously for 20 seconds.
3. Remove lateral flow device from foil pouch. Squeeze 2 drops of extract onto sample well on lateral flow device.
4. Wait until blue control line appears and then read result (about 2 minutes). control line plus blue line at test window - positive test. Control line only - negative result.

On-site detection of Phytophthora, Pythium and Rhizoctonia in
15 minutes.

ALERT - Summary User Guide
1. Collect samples and grind with Extrak pads (provided).
2. Insert Extrak strips in extraction solution in bottle 1. Shake 20 times.
3. Break off 3 od antibody pre-coated microtitre wells. Place in well holder.
4. Add 3 drops positive control to first well, 3 drops negative control to second well and 3 drops sample to third well. Incubate 5 minutes.
5. Empty wells and wash 5 times with water using wash bottle provided.
6. Add 3 drops of antibody + conjugate to each well. Incubate 5 minutes.
7. Repeat step 5.
8. Add 3 drops of substrate to all wells. Incubate 5 minutes. Do not wash out wells.
9. Add 3 drops Red Stop Solution and shake gently to mix.
10. Read result visually by comparing sample well colour with positive and negative well colours

ADGEN FLUORISCAN IF ^TM
Detection of bacteria by imunofluorescence.

ADGEN FLUORISCAN IF - User Guide
1. Extract sample by grinding plant tissue in buffer.
2. Shake positive and negative controls to resuspend cells.
3. Place 20μl drop of sample extract to one well on slide provided. Repeat for each control.
4. Add 20μl acetone to each drop. Leave 30 minutes. This fixes bacteria to slide.
5. Wash slide with buffer (PBS).
6. Add 20μl of specific antibody diluted as recommended to each spot on slide. Leave 30 minutes.
7. Wash slide with buffer (PBS).
8. Add 20μl of FITC antibody diluted as recommended to each spot on slide. Leave 30 minutes.
9. Wash slide with buffer (PBS).
10. Mount slide in buffered glycerol and examine under UV microscope.

SPOT CHECK - User Guide
1. 1. Grind plant tissue in disposable grinder provided.
2. 2. Attatch test tip to syringe provided.
3. 3. Draw extract into test tip and expel 5 times. Incubate for 5 minutes.
4. 4. Expel sap from test tip.
5. 5. Draw into test tip liquid from well 1 in test tray. Incubate for 5 minutes.
6. 6. Expel liquid from test tip.
7. 7. Repeat 4 & 5 with liquid from well 2 in test tray.
8. 8. Draw liquid from well 3 in test tray through test tip and into syringe. Do not expel.
9. 9. Draw liquid from well 4 in test tray into test tip. Incubate for 5 minutes.
10. 10. Stop reaction by expelling contents of syringe through test tip.
11. 11. Examine tip for colour development. Colour present - positive test. No colour - negative test.
